Tato diplomová práce se zabývá problematikou mnohočetné lékové rezistence (MDR) nádorových buněk vůči inhibitorům HSP90. Nádorové buňky s MDR fenotypem vyka-zují sníženou senzitivitu proti lékům, které jsou strukturně a funkčně odlišné. V teoretické části diplomové práce jsou shrnuty klasické mechanismy mnohočetné lékové rezistence se zaměřením na ABC transportéry a proteiny tepelného šoku. V experimentální části byly získány rezistentní linie buněk vůči geldanamycinu, což je specifický inhibitor HSP90. Tyto rezistentní buňky byly dále charakterizovány a byly analyzovány mechanismy jejich získané rezistence. Zaznamenali jsem, že rezistence buněk A549 vůči geldanamycinu je pravděpodobně spjata se zvýšenou expresí ABCC1, jednoznačné důkazy pro tento závěr ovšem nebyly získány. Lze vyloučit, že by rezistence byla spojena se zvýšenou expresí ABCB1, ABCG2 nebo HSP90, HSP70, HSP27.

This thesis deals with the issue of multidrug resistance (MDR) of tumor cells to HSP90 inhibitors. Tumor cells with an MDR phenotype show reduced sensitivity against drugs that are structurally and functionally different. The theoretical part of the thesis sum-marizes the classic mechanisms of multiple drug resistance with a focus on ABC trans-porters and heat shock proteins. In the experimental part, cells resistant to geldanamy-cin, which is a specific inhibitor of HSP90, were obtained. These resistant cells were further characterized, and the mechanisms of their acquired resistance were analyzed. We note that the resistance of A549 cells to geldanamycin is likely to be associated with increased expression of ABCC1, but unequivocal evidence for this conclusion has not been obtained. It can be ruled out that resistance is associated with increased ex-pression of ABCB1, ABCG2 or HSP90, HSP70, HSP27.
