Bakalářská práce se zabývá problematikou interpretace variantami nejasného významu nalezených během testování pacientů s podezřením na dědičný nádorový syndrom. A jejích interpretací v kontextu nově získaných informací o frekvenci variant v genomu české populace analyzovaném v projektu A-C-G-T (Analýza českých genomů pro teranostiku). Hereditární nádorové syndromy tvoří 5-10% všech nádorových onemocnění. Mezi nejčastěji se vyskytující dědiční nádorová onemocnění patří karcinom prsu, vaječníků, kolorekta a dělohy. Metody masivního paralelního sekvenování umožňují rychlou identifikaci klinicky významných variant. V praktické části byla provedena analýza 815 vzorků v letech 2018-2022. Celkem bylo nalezeno 164 variant nejasného významu u pacientů testovaných na hereditární nádorové syndromy. Cílem praktické části bakalářské práce bylo seznámení se s metodou NGS pomocí analýzy vzorků DNA získané z periferní krve pacientů s různými nádory a podezřením z hereditární nádorové predispozice a následná analýza 164 nalezených variant nejasného významu pomocí porovnávání s údaji získanými v projektu A-C-G-T a mezinárodní populační databází gnomAD.

The bachelor thesis deals with the problem of interpretation of variants of unclear significance found during testing of patients with suspected hereditary cancer syndrome. And its interpretation in the context of newly obtained information on the frequency of variants in the genome of the Czech population analysed in the A-C-G-T project (Analysis of Czech genomes for theranostics). Hereditary cancer syndromes account for 5-10% of all cancers. Breast, ovarian, colorectal and uterine cancers are the most common hereditary cancers. Massively parallel sequencing methods allow rapid identification of clinically relevant variants. In the practical part, 815 samples were analyzed between 2018 and 2022. A total of 164 variants of unclear significance were found in patients tested for hereditary cancer syndromes. The aim of the practical part of the bachelor thesis was to learn about the NGS method by analyzing DNA samples obtained from peripheral blood of patients with various cancers and suspected hereditary cancer predisposition and then analyzing the 164 found variants of uncertain significance by comparing them with data obtained in the A-C-G-T project and the international population database gnomAD.
