Lékové rezistence jsou jedním z významných faktorů, které komplikují léčbu onkologických pacientů. V rámci této práce byla provedena příprava nádorových linií rezistentních vůči známým léčivům (cytarabin, fludarabin, 6-thioguanin, 6-merkaptopurin, cisplatina, karboplatina), ale také vůči experimentálním látkám (roskovitin, PNH173, PNH192, AB-61, CuEt). V případě látek s nukleosidovou strukturou byla příprava úspěšná a byly získaný linie s výrazně rezistentním fenotypem. U těchto linií byl pozorován nejen rozvoj na látku, která byla využita k samotné selekci, také zkřížená rezistence s dalšími látkami. V případě selekce cisplatinou a karboplatinou nebyl rozvoj rezistence tak výrazný a v případě látky CuEt k nárůstu rezistence prakticky nedošlo. V rámci základní charakterizace těchto linií byla ověřena exprese dvou proteinů spojených s mnohočetnou lékovou rezistencí, P-glykoproteinu a Multidrug resistance-associated proteinu 1 (MRP1). Dále pak byla ověřena míra exprese nukleosidových membránových transportérů (ENT, CNT) a vybrané linie byly použiti pro screening nových látek, které by mohly sloužit jako potenciální protinádorová léčiva.

Drug resistance is one of the key problems in cancer therapy. In this thesis, we were mainly focused on the development of cell lines resistant to known drugs (cytarabine, fludarabine, 6-thioguanine, 6-mercaptopurine, cisplatin, carboplatin) and also to experimental compounds (roscovitine, PNH713, PNH192, AB-61, CuEt). Selection done by nucleoside-based compounds was successful for all compounds and we detected secondary and also cross resistant phenotype. Selection by cisplatin and carboplatin did not lead to development of such a huge increase in resistant phenotype compared to nucleosides and in the case of CuEt selection, there was almost no change in the sensitivity after the selection. Basic characterization was done for all selected cell lines, including expression of MDR proteins, such as P-glycoprotein a Multidrug resistance-associated protein and expression of nucleoside membrane transporters (ENTs, CNTs). Some of the resistant cell lines were used for the screening of new compounds with promising anticancer activity.
