Cytokininy jsou rostlinné hormony ovlivňující růst a vývoj rostlin. Strukturně se jedná o N6 deriváty adeninu. Cytokininy (CK) jsou syntetizovány dvěma možnými způsoby. Prvním způsobem je biosyntéza de novo, která spočívá v prenylaci adeninu za vzniku isopentenyladeninu nebo trans-zeatinu. Druhým způsobem je tzv. nepřímá biosyntéza, která spočívá prenylaci adeninu, který je obsažen v transferové RNA (tRNA), následně při její degradaci jsou cytokininy cis-zeatinového typu uvolněny do okolí. Cytokininy jsou v rostlině transportovány xylémem do nadzemní části rostliny a floémem zpět ke kořenům. Transport probíhá pomocí transportérů. Mezi transportéry CK patří Purinové permeasy (PUP) ,které transportují nukleobáze, dále Ekvilibrativní nukleosidové transportéry (ENT) transportující nukleosidy. Dalším transportérem je ABCG14, který se podílí na transportu na dlouhé vzdálenosti. Nedávno objevený transportér AZG2, který hraje důležitou roli při vývoji laterálních kořenů. Cílem praktické části bakalářské práce byly selekce homozygotních rostlin vybraných T-DNA inzerčních linií Arabidopsis thaliana v genech, které kódují CK transportéry rodiny ENT a transportér ABCG14. toho bylo docíleno pomocí PCR genotypizace a následné vizualizace pomocí elektroforézy. Homozygotní linie rostlin budou následně použité v dalším výzkumu.

Cytokinins are plant hormones that affect plant growth and development. Structurally, they are N6 derivatives of adenine. Cytokinins (CK) are synthesized in two possible ways. The first way is de novo biosynthesis, which involves the prenylation of adenine to form isopentenyladenine or trans-zeatin. The second way is the so-called indirect biosynthesis, which consists in the prenylation of adenine contained in transfer RNA (tRNA), then upon its degradation, cis-zeatin-type cytokinins are released into the environment. Cytokinins are transported in the plant by the xylem to the aerial part of the plant and by the phloem back to the roots. Transport is by transporters. CK transporters include Purine permeases (PUPs) that transport nucleobases, and Equilibrative nucleoside transporters (ENTs) that transport nucleosides. Another transporter is ABCG14, which is involved in long-distance transport. A recently discovered transporter, AZG2, plays an important role in lateral roots development. The main goal of the practical part of the bachelor thesis was to select homozygous plants of selected T-DNA insertion lines of Arabidopsis thaliana in genes encoding CK transporters of the ENT family and ABCG14 transporter. This was achieved by PCR genotyping and subsequent visualization by electrophoresis. The homozygous plant lines will subsequently be used in further research.
