The diploma thesis is focused on hyperthermic treatment, especially with mild temperatures, and its use in cancer therapy. The thesis deals with approaches, how to reach hyperthermia treatment in patients, mechanisms of action, and possibilities of hyperthermia usage as adjuvant therapy with other treatment modalities. We discuss the problematic aspects of applying high non-physiological temperatures and the advantages of long-term mild hyperthermia. The thesis underlines the rationale for the utilization of hyperthermia in cancer therapy.
The experimental part of the thesis reports the effect of long-term mild hyperthermia on various cancerous and non-cancerous cell lines. These comprise evaluating cell proliferation, a cytotoxic effect on cells, stress signalling, heat shock protein response, DNA damage response, detection of replicating cells, and cell cycle analysis. The results show a cytostatic and also cytotoxic effect of long-term mild hyperthermia for numerous cancerous cell lines, whereas non-cancerous cells are more resistant to this treatment. In addition to this, acquired data demonstrate the capability of long-term mild hyperthermia to potentiate the effect of the genotoxic agent olaparib.

Cancer is one of the leading causes of mortality worldwide. The common alternatives used to treat cancers can be classified into conventional and modern methods. Surgery, radiation therapy, and chemotherapy are considered as the three standard methods for cancer treatment but suffer from many limitations. Hyperthermia has been recently introduced as adjuvant therapy for cancer and holds great promise for combating this disease (1-4). Hyperthermia is defined as a treatment in which the targeted tissue is exposed to high temperatures that either destroy the cancer cells directly or sensitize them to other treatment modalities (5,6). The preferential sensitivity of cancer cells to thermal therapy arises from their naturally elevated level of abnormal proteins, which are easily attacked by thermal damage, leading to protein unfolding, aggregation, and/or denaturation causing proteotoxic stress (7). This work will aim to study so-called long-term mild hyperthermia. In some aspects, such a treatment modality follows up on successes of Coley´s method for cancer treatment using purposely induced fever (8). Nowadays long-term mild hyperthermia of target tissues in patients can be theoretically achieved by portable battery-powered electric field generators eliminating problematic aspects of systemic fever however the studies on the efficacy of such therapeutical options are missing. In this work, the student will test in various cancer cell lines how long-term hyperthermia affects cell growth and the cytotoxic effect of standard chemotherapy. In addition to this, the activation of heat shock protein response, protein turnover, and pro-apoptotic signaling pathways will be evalutaed using western blot and immunofluorescence techniques.
