Diplomová práce se věnuje roli proteinového komplexu P97/NPL4 a jeho zapojení do opravy poškozené DNA. Literární přehled je zaměřen na ubikvitin-proteazomový systém a jeho nezbytnou složku, segregázu P97/VCP. P97 je důležitý regulátor proteostázy, ale v posledních letech se výzkum zaměřuje také na jeho roli při opravě DNA. Jeho kofaktory NPL4-UFD1 obsahují motiv rozpoznávající ubikvitin, čímž umožňují segregáze vázat polyubikvitinované substráty. Těmi mohou být také polyubikvitinované proteiny navázané na poškozenou DNA. Experimentální část práce je zaměřena na metodu laserové mikroiradiace, pomocí níž byly v buněčných liniích stabilně exprimujících GFP-značené proteiny indukovány léze v DNA a poté byla v čase pozorována vazba těchto proteinů do míst poškození. Následně byl studován vliv různých inhibitorů ubikvitin-proteazomového systému Bortezomibu, CB-5083, MLN7243 a CuET na shlukování těchto proteinů v místech lézí. Zároveň byla testována kombinace inhibitoru P97 CB-5083 s ionizujícím zářením a látkami indukujícími poškození DNA jako je cisplatina, doxorubicin a inhibitor ATR kinázy AZD6738.

This diploma thesis deals with the topic of the protein complex P97/NPL4 and its involvement in the repair of the damaged DNA. The literary review is focused on the ubiquitin-proteasome system and its essential component, segregase P97/VCP. P97 is an important regulator of proteostasis, however, the research also focuses on its involvement in the repair of the damaged DNA. Its cofactors NPL4-UFD1 contain a ubiquitin-recognizing motif, which allows segregase to bind itself on the polyubiquitinated substrates. These substrates may represent the
polyubiquitinated proteins bonded on the damaged DNA. The experimental part of the thesis is focused on the method of laser microirradiation, which allows precise induction of DNA lesions. Cell lines expressing various GFP-tagged proteins were microirradieated and followed in time by confocal microscopy to monitor the recruitment of GFP- reporters. Moreover, various inhibitors of the ubiquitin-proteasome system like Bortezomib, CB-5083, MLN7243 and CuET were used to investigate the determinants of the accumulation of these proteins in the sites of damage. The combinations of the inhibitor P97 CB-5083 with ionizing radiation and DNA damage-inducing agents like cisplatin, doxorubicin and AZD6738, an inhibitor of ATR kinase, were also tested.
