Předkládaná diplomová práce se věnuje pochopení mechanismů interakce signálních fototropinových a stresových drah probíhajících v modelové rostlině Solanum lycopersicum L. Teoretická část byla věnována aktuální problematice abiotického stresu a světelných signálních drah modelových rostlin. Experimentální část zahrnuje analýzu exprese vybraných kandidátních genů PHOT1 a PHOT2 v semenech rajčete jedlého a mutantního genotypu 7B-1, vyznačujícího se zvýšenou tolerancí k abiotickým stresům na modrém světle. Ke sledování exprese byla použita metoda kvantitativní PCR. Výsledky prováděných experimentů vedly k závěru, že modré světlo a funkční produkt 7B-1 může zvyšovat toleranci semen rajčete k zasolení prostřednictvím fotoreceptoru PHOT1 a PHOT2.

This diploma thesis deals with the understanding of the mechanisms of cross-talk between phototropin and stress signalling pathways in the model plant Solanum lycopersicum L. The theoretical part was devoted to current problems of abiotic stress and light signalling pathways in the model plants. The experimental part includes the analysis of the expression of selected candidate genes PHOT1 and PHOT2 in tomato seeds of the edible and mutant genotype 7B-1, characterized by enhanced tolerance to abiotic stress in blue light. Quantitative PCR was used to monitor the gene expression. The results of
the conducted experiments led to the conclusion that the blue light and the functional product 7B-1 increase the tolerance of tomato seeds to salinization through the photoreceptor PHOT1 and PHOT2.
