Organokovové komplexy sú významnou súčasťou protinádorového výskumu od úspešného klinického schválenia cisplatiny. Popri konvenčných platinových cytostatikách, ktoré však majú svoje limity, je dôležitý výskum a vývoj v tejto oblasti za účelom získania nových koordinačných komplexov s centrálnym atómom kovu s lepším terapeutickým účinkom a nižšou systematickou toxicitou. Predkladaná dizertačná práca sa zaoberá mechanistickým štúdiom protinádorového účinku troch skupín nových platinových zlúčenín na molekulárnej úrovni a štúdiom komplexu gália na bunečnej úrovni.
Prvá časť práce pojednáva o výsledkoch štúdia in vitro interakcií nových fotoaktivovateľných dinukleárnych platičitých komplexov s DNA a porovnaním ich účinku a zároveň vplyvu dĺžky spojovacieho linkru s príbuzným mononukleárnym FM190 (trans, trans, trans- [Pt(N3)2(OH)2(pyridín)2]). Zistilo sa, že po ožiarení modrým svetlom dochádza k fotodekompozícii študovaných dinukleárnych komplexov za vzniku dvoch aktívnych mononukleárnych platinových jednotiek podobných mononukleárnemu FM190. Následkom toho spôsobovali skúmané dinukleárne komplexy takmer rovnaké poškodenie DNA ako mononukleárny FM190.
Výsledky prezentované v druhej časti tejto dizertačnej práce ukazujú, že chiralita komplexov [Pt(DABN)Cl2], kde (DABN = 1,1-binaftyl-2,2-diamín), môže výrazne ovplyvniť spôsob ich väzby na DNA, ktorá sa všeobecne považuje za hlavný farmakologický cieľ konvenčných protinádorových komplexov platiny a ich derivátov. Na druhej strane sa však rozdiely vo väzbe na DNA neprenášajú priamo do bunečného spracovania a toxicity R- a S-[Pt(DABN)Cl2 v rakovinových bunkách. Z toho vyplýva, že poškodenie DNA týmito komplexami nemusí byť hlavným dôvodom ich protinádorového účinku na rozdiel od konvenčných platinových cytostatík a ich derivátov.
Treťou časťou práce je štúdium komplexu gália na bunečnej úrovni a stanovenie jeho antiproliferatívnej aktivity na viacerých bunkových nádorových líniách. V porovnaní s cisplatinou je tento komplex výrazne účinnejší a selektívnejší na nádorové bunky.
Jednou z príčin rezistencie na liečbu je tolerancia poškodenia DNA prostredníctvom translézovej syntézy DNA cez platinové adukty. V poslednej časti tejto dizertačnej práce bola študovaná translézová syntéza DNA cez špecifický monofukčný adukt protinádorového komplexu AMD ([PtCl(en)(L)](NO3)2 en = ethylén-1,2-diamín, L = N-[2-(akridín-9-ylamino)ethyl]-N-methylpropionamidín)) pomocou DNA polymeráz a s tým spojené termodynamické zmeny prostredníctom mikroškálovej termoforézy (MST). Výsledky získané pomocou MST boli v súlade s experimentálnymi dátami popisujúcimi translézovú syntézu DNA cez adukt AMD prokaryotickou aj eukaryotickou DNA polymerázou. Termodynamické parametre taktiež prispeli k hlbšiemu porozumeniu toho, ktoré faktory vplývajú na mechanizmus syntézy DNA cez adukt AMD.

Organometallic complexes have been an important part of anticancer research since the successful clinical approval of cisplatin. In addition to conventional platinum cytostatics, which have limitations, research and development in this field are essential to obtain new metal - based coordination complexes with a better therapeutic effect and lower systemic toxicity. The present dissertation thesis deals with the mechanistic study of the antitumor effect of three groups of new platinum compounds at the molecular level and the study of the gallium complex at the cellular level.
The first section pertains to the results of an in vitro study of DNA interactions of new photoactivatable dinuclear platinum (IV) complexes as well as the comparison of their effects and the influence of the length of the linker with the related mononuclear FM190 (trans, trans, trans- [Pt(N3)2(OH)2(pyridine)2]). It was found that after irradiation by blue light, photodecomposition of the studied dinuclear complexes occurred to form two active mononuclear platinum (II) units similar to mononuclear FM190. As a consequence, the investigated dinuclear complexes caused almost the same DNA damage as mononuclear FM190.
The results presented in the second section of this dissertation show that the chirality of [Pt(DABN)Cl2 (DABN = 1,1-binaphtyl-2,2-diamine) complexes can significantly affect the way they bind to DNA, which is commonly considered the main pharmacological target of conventional antitumor platinum complexes and their derivatives. On the other hand, the variations in DNA binding do not translate directly into cellular processing and toxicity of R- and S-[Pt(DABN)Cl2 in cancer cells. Therefore, unlike conventional platinum drugs and their derivatives, the DNA damage resulting from binding of R- and S-[Pt(DABN)Cl2 is not the major factor responsible for their biological actions.
The third section of this thesis describes the study of the gallium complex at the cellular level and the determination of its antiproliferative activity on several tumor cell lines. Compared to cisplatin, this complex is significantly more effective and selective for tumor cells.
One cause of resistance to treatment is the tolerance of DNA damage by translesion DNA synthesis through platinum DNA adducts. The last section of this dissertation deals with the study of the translesion DNA synthesis through site-specific monofunctional adduct formed by antitumor AMD complex [PtCl(en)(L)](NO3)2, en = ethane-1,2-diamine, L = N-[2-(acridin-9-ylamino)ethyl]-N-methylpropionamidine) using DNA polymerases and MicroScale thermophoresis (MST) for associated thermodynamic changes. The MST results were in good agreement with experimental data describing translesion DNA synthesis across the AMD adduct by DNA polymerases of eukaryotic and prokaryotic origin. The equilibrium thermodynamic data also contributed to a deeper understanding of the factors that control the mechanisms of template-directed DNA synthesis across the AMD adduct.
