Táto dizertačná práca je zameraná na štúdium chemického zloženia osemenia hrachu siateho (Pisum sativum) a interpretáciu získaných dát v kontexte fyzikálnej dormacie semien. Okrem toho bola zameraná pozornosť aj na štúdium chemických zmien v osemení hrachu siateho po simulácii teplotného cyklovania semien.
Úvod dizertačnej práce v krátkosti poukazuje na význam dormancie semien v prírode a jej nežiadúce pôsobenie v poľnohospodárstve. V teoretickej časti sú zhrnuté techniky, ktoré boli do dnešnej doby využité v analýze rastlinných pletív. Podrobnejšie sú diskutované metódy, ktoré boli zamerané na analýzu rastlinných pletív v osemení. V tejto časti je tiež prehľadne predstavená stavba osemenia u strukovín a problematika fyzikálnej dormacie a jej regulácie (zameranej najmä na vplyv teploty na chemické zmeny v osemení).
V experimentálnej časti boli vyvinuté metódy pre analýzu povrchov osemení hrachu založených na hmotnostnej spektrometrii s (matricou asistovanou) laserovou desorpciou/ionizáciou ((MA)LDI-MS) a s ionizáciou priamou sondou pre analýzu pevných látok za atmosferického tlaku (ASAP-MS). Boli optimalizované experimentálne parametre a preštudované možnosti využitia iónovo mobilitnej separácie pre identifikáciu látok. Tieto metódy boli aplikované na analýzu semien šiestich vybraných odrôd hrachu siateho, troch dormantných (D) a troch nedormantných (N). Jednalo sa o dormantné (divoké) semená druhu Pisum sativum subsp. elatius (genotypy JI64, VIR320, L100) a semená nedormantného (kultúrneho) hrachu Pisum sativum subsp. sativum (genotypy JI92, Cameor, Terno). Následne boli experimenty rozšírené aj o analýzu RIL línií (rekombinantne krížené línie) hrachu siateho, získané vzájomným krížením genotypov JI64 a JI92. Pre štúdium distribúcie analytov na povrchu osemení bolo využité aj hmotnostno spektrometrické zobrazovanie. ASAP-MS bola na priamu analýzu povrchu semien strukovín použitá prvýkrát. (MA)LDI-MS a ASAP-MS v kombinácii s viacrozmernými štatistickými metódami, PCA a OPLS-DA, umožnili meraním osemení zrelých semien klasifikáciu (rozlíšenie) dormantných a nedormantných genotypov hrachu. Týmito technikami boli nájdené markery so signifikantným signálom pre dormantné genotypy hrachu, a to hydroxylované a nasýtené vyššie mastné kyseliny.
V závere experimentálnej časti je riešená problematika zameraná štúdium chemických zmien v osemení hrachu po simulovanom teplotnom cyklovaní (periodické zmeny znižovania a zvyšovania teploty, konkrétne na teploty 25 a 60°C). V tejto štúdii bola pozornosť zameraná na dva vybrané genotypy T14/5 (číselný kód 86) a P016 (číselný kód 17), ktoré boli analyzované pomocou techník infračervenej spektroskopie (ATR-FTIR) a hmotnostnej spektrometrie s využitím laserovej desorpsie/ionizácie (LDI-MS) a tiež v spojení s kvapalinovou chromatografiou (LC/ESI-MS). Bolo zistené, že genotypy po teplotnom cyklovaní variujú v obsahu niektorých látok oproti svojim kontrolám. V genotype 86 bol detekovaný vyšší obsah galokatechínu a diméru galokatechínu po teplotnom cyklovaní. Pri genotype 17 bol po cyklovaní pozorovaný pokles obsahu saponínu (chromosaponínu I). Tieto látky sa môžu podieľať na regulácii dormancie u týchto semien.
Techniky LDI-MS a ASAP-MS predstavujú užitočný nástroj v priamej analýze rastlinného materiálu a umožňujú detailnú analýzu profilu mastných kyselín v povrchových vrstvách rastlinných pletív bez nutnosti extrakcie semena rozpúšťadlami, ktorá je pracná a môže byť zdrojom chýb (napr. vznikom nežiadúcich artefaktov, malým extrakčným výťažkom apod.). Veľkou výhodou vyvinutej ASAP-MS techniky je možnosť analyzovať zloženia osemenia na celých kompaktných a neporušených semenách.

This dissertation thesis is focused on the study of the chemical composition of Pisum sativum and on the interpretation of obtained data in the context of physical dormancy of seeds. In addition, the attention was also focused on the investigation of chemical changes in pea seeds after temperature cycling simulation.
The importance of seed dormancy in the nature and its undesirable features in agricultural field is described in the Introduction. The techniques that have been performed to date in the analysis of plant tissues are summarized as well. Techniques used in the seed coat analysis are also discussed in more details. In this part, the structure of seed coat from legumes and the issue of physical dormancy and its regulation (including the influence of temperature) are also presented.
In the experimental part, methods for the analysis of seed coat surface utilizing mass spectrometry with (matrix assisted) laser desorption / ionization ((MA)LDI-MS) and Atmospheric Solid Analysis Probe (ASAP-MS) were developed. The experimental parameters were optimized and the possibilities of using ion mobility separation for marker identification were investigated. These methods were applied to the analysis of seeds from six selected varieties of pea, three dormant (D) and three nondormant (N). Dormant (wild) seeds Pisum sativum subsp. elatius (genotypes JI64, VIR320, L100) and nondormant (cultivated) seeds Pisum sativum subsp. sativum (genotypes JI92, Cameor, Terno) were studied. Subsequently, the experiments were extended by analysis of RIL lines (recombinant cross lines) of pea, obtained with breeding of JI64 and JI92 genotypes. Mass spectrometric imaging was used to study the distribution of analytes on the seed coat surface. ASAP-MS was used for the direct analysis of legume seed coat surface for the first time. (MA)LDI-MS and ASAP-MS in combination with multidimensional statistics, PCA and OPLS-DA, allowed to classify dry and mature seeds with respect to dormancy (separate them into dormant and nondormant group). These techniques revealed markers with significant signal for dormant pea genotypes, namely hydroxylated and saturated very long chain fatty acids.
Besides, chemical changes in the pea seed coats after thermal cycling were investigated. In this study, the attention was focused on genotype T14/5 (code number 86) and P016 (code number 17), that were analyzed using infrared spectroscopy (ATR-FTIR), laser desorption/ionization mass spectrometry (LDI-MS) and liquid chromatography/mass spectrometry (LC/ESI-MS). It was revealed that genotypes after thermal cycling vary significantly in the content of some substances compared to their controls. In genotype 86, a higher content of gallocatechin and gallocatechin dimer was detected after thermal cycling. In genotype 17, a decrease in saponin (chromosaponin I) content was observed after thermal cycling. These substances may be involved in the regulation of dormancy in these seeds.
LDI-MS a ASAP-MS techniques represent a useful tool in direct analysis of plant material and allow detailed analysis of the fatty acid profile in the surface layers of plant tissues without the solvent extraction of the seeds, that is laborious and can be a source of errors (e.g. a great advantage of the developed ASAP-MS technique is the ability to analyze the composition of the seed coat on intact seeds).
