Tato bakalářská práce se zabývá syntézou nových N-nitro a N-aminobenzyl derivátů pyrrolochinolinu působících prostřednictvím interakce s kolchicinovým vazebným místem jako selektivní inhibitory polymerace tubulinu, čehož je využíváno především v protinádorové terapii. Součástí této práce je literární rešerše zabývající se klasifikaci derivátů pyrrorochinolinu vykazujících protinádorové účinky, jejich vlastnostmi a využitím. Výsledky a diskuse a dále experimentální část práce se zabývají optimalizaci reakčních podmínek vedoucích k novým derivátům 4-chlorpyrrolo[3,2 c]chinolinu nesoucích N-nitrobenzylovou a N-aminobenzylovou skupinu. Připravené deriváty byly podrobeny cytotoxickému a antimikrobiálnímu screeningu vůči vybraným nádorovým buňkám a bakteriálním kmenům. Na závěr byly tyto deriváty podrobeny studiu vztahů mezi jejich strukturou a biologickou aktivitou pomocí in silico modelů a experimentálních dat a získané výsledky byly porovnány s dříve připravenými deriváty pyrrolochinolinu.

This bachelor's thesis deals with the synthesis of novel pyrroloquinoline derivatives acting through interaction with the colchicine binding site as selective inhibitors of tubulin polymerization, which is the strategy used in antitumor therapy. First, the literature research is provided with the focuses on classification of pyrroloquinoline derivatives acting as antitumor agents, their properties and use. The chapter Results and discussion and following Experimental part of this work describe optimization of reaction conditions leading to novel 4-chloropyrrolo[3,2-c]quinoline derivatives bearingN-nitro and N-aminobenzyl groups. The final products were subjected to test of their cytotoxic and antimicrobial activity against selected cytotoxic cells and bacterial strains. Finally, in silico model and relationship between their structure and biological activity were studied. Last but not least, the obtained in silico and experimental data were evaluated and compared with the previously obtained data for pyrroloquinoline derivatives.
