Diplomová práce charakterizuje pšenici jako významnou plodinu pro výživu lidstva. Zdůvodu vysokého zastoupení obilnin vosevních postupech se houbové choroby stávají stále větší hrozbou pro produkci pšenice.
Nejvýznamnější chorobou pat stébel pšenice je stéblolam způsobovaný houbami Oculimacula yallundae a Oculimacula acuformis. Jedná se o patogeny způsobující vysoké ztráty na výnosech se stále zvyšující se incidencí. Fungicidní ochrana proti stéblolamu je účinná, ale u řady látek dochází kjejich zákazu a patogen při opakovaném ošetření stejnou účinnou látkou získává rezistenci. Ztohoto důvodu je nezbytné šlechtění rezistentních odrůd pšenice, které snižuje riziko vzniku rezistence patogenů a nutnost použití chemické ochrany rostlin.
Mnohé rezistentní odrůdy nesou gen rezistence Pch1, který se vmnoha výzkumech osvědčil jako velmi účinný. Experimenty této diplomové práce testovaly odrůdy nesoucí tento gen a další odolné odrůdy bez tohoto genu. Pomocí qPCR byly vrostlinných pletivech zaznamenány oba druhy patogenů, kvantitativní hodnoty byly srovnány svizuálními příznaky a výsledky byly použity pro srovnání tolerance a rezistence testovaných odrůd.
Nakonec vsouvislosti sdiagnostikou patogenů byly vtéto diplomové práci srovnány metody vizuálního hodnocení a molekulární metody pro zjištění odolnosti odrůd, diagnostiku patogenů a vliv klimatických podmínek.
Annotation in English
The thesis characterizes wheat as an important crop for the nutrition of mankind. Due to the high proportion of cereal crops in agricultural production practices, fungal diseases are becoming an increasing threat to wheat production.
The most important disease of wheat stalk pathogen is eyespot caused by the fungi Oculimacula yallundae and Oculimacula acuformis. These are pathogens causing high yield losses with increasing incidence. Fungicidal protection against eyespot is effective, but many substances are being banned and the pathogen acquires resistance when treated repeatedly with the same active substance. For this reason, the breeding of resistant wheat varieties is necessary to reduce the risk of pathogen resistance and the need for chemical plant protection.
Many resistant varieties carry the Pch1 resistance gene, which has proven to be very effective in many studies. The experiments in this thesis tested varieties carrying this gene and other resistant varieties without this gene. Using qPCR, both types of pathogens were recorded in plant tissues, quantitative values were compared with visual symptoms and the results were used to compare tolerance and resistance of the tested varieties.
Finally, in the context of pathogen diagnosis, visual assessment and molecular methods were compared in this thesis to determine varietal resistance, pathogen diagnosis and the effect of climatic conditions.
