Přechod od oocytu k embryu (oocyte-to-embryo transition, OET) je klíčovým procesem v reprodukci savců. V období meiotického zrání a raného emryonálního vývoje, kdy je utlumena transkripce, je pro správný vývoj vyžadována precizní kontrola translace, což se děje prostřednictvím proteinů vážících se na RNA. Cílem této diplomové práce bylo prozkoumat roli CPEB3 proteinu ve vývoji oocytu a raného embrya. Výsledky ukázaly, že nepřítomnost CPEB3 negativně ovlivňuje ženskou plodnost. Oocyty bez CPEB3 bez potíží dokončí meiozu, ale ve fázi 2 buněk výsledného embryo se vývoj zastaví následkem deregulovanému transkriptomu, který vedl k expresi nesprávného proteomu, který není schopen aktivovat embryonální transkripci (ZGA). Tato studie dale ukázala, že CPEB3 reguluje translaci určité podskupiny transkriptů prostřednictvím kontroly jejich stability a polyadenylace. Celkově tato práce identifikovala CPEB3 jako klíčový maternální faktor, který předurčuje vývojovou kompetenci oocytu.

This diploma thesis investigates the role of cytoplasmic polyadenylation element-binding protein 3 (CPEB3) in oocyte and early embryonic development. The results showed that the absence CPEB3 negatively affects female fertility. CPEB3-depleted oocytes undergo meiosis normally, but they experience developmental arrest due to impaired transcriptome, leading to aberrant protein expression and subsequent failure of the embryonic transcription onset. In addition, this study demonstrates that CPEB3 promote translation of subset of mRNAs through control their stability and polyadenylation. Taken together, this thesis identified CPEB3 as a key maternal factor that determines oocyte developmental competence.

This work provides a strategy which will be beneficial for unveiling the mechanism of CPEB3 action during oocyte-to-embryo transition and explaining the 2cell embryonic arrest and subsequent infertility observed in CPEB3 cKO mice.
1. qRT-PCR validation of the most significant and phenotype-related genes from RNA-Seq datasets and following Western blot analysis. The expression analysis will be performed on CPEB3 cKO GV and MII oocytes and 2-cell embryos using heterozygotes as controls.
2. The poly(A) length of these transcripts will be determined via PolyAdenylation Tailing assay (PAT) in GV and MII oocytes and 2cell embryos to detect supposed changes between wild type and cKO. The comparison between these two groups will be performed to test the hypothesis that CPEB3 is part of the regulatory network controlling mRNA stability and translation via polyadenylation.
3. To prove the role of CPEB3 in polyadenylation of selected mRNAs, the samples will be treated with cordycepin to block poly(A) lengthening (Fuchimoto et al. 2001), expecting that a) treated WT oocytes and zygotes will mimic the polyA status and expression level in the untreated cKO samples and furthermore leads to observed phenocopy and b) no differences between transcripts in treated and untreated cKO will be observed.
