Tato diplomová práce se zabývá antiproliferační aktivitou derivátů oxysterolů in vitro. Oxysteroly jsou bioaktivní látky, které vznikají oxidací molekuly cholesterolu, a mnoho z nich má významné cytotoxické a proapoptotické vlastnosti, které by mohly nalézt uplatnění v protinádorové terapii. V teoretické části práce jsou shrnuty informace o cervikálním karcinomu, buněčném cyklu, programované buněčné smrti a oxysterolech. V experimentální části práce bylo provedeno testování antiproliferační aktivity dvou syntetických a dvou přírodních derivátů oxysterolů na buněčné linii HeLa odvozené od nádoru děložního hrdla. Metodou western blottingu byly sledovány hladiny proapoptotických proteinů a jaderného receptoru. Průběh buněčného cyklu byl analyzován pomocí průtokové cytometrie a metodou imunofluorescence byla sledována translokace receptoru do jádra buněk.

The diploma thesis deals with antiproliferative activity of oxysterol derivatives in vitro. Oxysterols are bioactive substances that arise from the oxidation of the cholesterol molecules, and many of them have significant cytotoxic and proapoptotic properties that could find application in antitumor therapy. The theoretical part of the thesis summarizes information about cervical cancer, the cell cycle, programmed cell death and oxysterols. In the experimental part of the thesis the antiproliferative activity of two synthetic and two natural oxysterol derivatives was tested on the HeLa cell line derived from cervical cancer. The levels of proapoptotic proteins and nuclear receptor were monitored by western blotting. The cell cycle progression was analyzed using flow cytometry and the nuclear receptor translocation was monitored using the immufluorescence method.
