Mitochondriální onemocnění jsou skupinou klinicky heterogenních poruch, které vznikají v důsledku dysfunkce elektronového transportního řetězce a oxidativní fosforylace. V současné době pro tyto poruchy neexistuje uspokojivá léčba.
Probíhá výzkum nových terapeutických přístupů jedním z nich je použití látek, které podporují biogenezi mitochondrií. Některé látky z knihovny naší laboratoře zvyšují signál v resazurinovém testu, aniž by zvyšovaly proliferaci lidských buněk. Možným vysvětlením je, že tyto látky indukují biogenezi mitochondrií. Optimalizovala jsem proto vybrané metody pro sledování mitogeneze. Množství mitochondriální masy jsem hodnotila jednak přímo pomocí automatické mikroskopie a analýzy obrazu, jednak nepřímo sledováním poměru DNA několika mitochondriálních lokusů a genomické DNA pomocí real-time PCR. Dále jsem sledovala aktivační fosforylaci AMP-aktivované kinázy spojenou s mitogenezí. Zjistila jsem, že indukce této fosforylace cytokininy je závislá
na buněčném typu.

Mitochondrial diseases are a clinically heteregenous group of disorders that arise as a result of dysfunction of the mitochondrial electron transport chain and oxidative phosphorylation. Currently, there are no satisfactory therapies
available. Novel therapeutic options are being investigated - one of them involves using agents enhancing mitochondrial biogenesis. Some compounds in our chemical library increase the signal in resazurin assay without promoting/stimulating human cells proliferation. This effect could result from induction of mitochondrial biogenesis. In this study, we optimized selected
methods for mitogenesis studies. The mitochondrial mass was quantified both directly by automated microscopy in connection with image analysis and indirectly by monitoring the ratio of several mitochondrial loci to genomic DNA
using real-time PCR. The activation phosphorylation of AMPactivated kinase associated with mitochondrial biogenesis was also evaluated.We report that induction of the phosphorylation by cytokinins depends on the cell type.

Cílem práce bude vyhodnotit vliv vybraných látek na biogenezi a funkci mitochondrií. Studován bude také vliv na viabilitu buněk a na jejich redoxní a energetický stav. Součástí práce bude rešerše shrnující stav vývoje farmakoterapie.
