Předložená disertační práce je zaměřena na vývoj a popis analytických metod stanovení vybraných biomarkerů v klinické diagnostice s využitím elektrochemie jako významného prvku moderních instrumentálních technik. Biomarkery jsou látky, jejich přítomnost může indikovat různé typy závažných onemocnění a stavů, či expozici škodlivým chemickým látkám. Pozornost byla v této práci věnována především stanovení metanefrinů, markerů tumoru feochromocytomu, a dále indoxyl sulfátu, významnému markeru řady zdravotních komplikací, od selhávání ledvin přes střevní dysbiózu až po poruchy centrální nervové soustavy. Část práce je též věnována vývoji originálního instrumentu sloužícího k metabolomickému výzkumu.
Spojení kapalinové chromatografie s elektrochemickým i spektrofotometrickým detektorem a propojení elektrochemie s hmotnostním spektrometrem byly využity s cílem analyzovat s vysokou citlivostí, přesností i správností vybrané sloučeniny ve složitých biologických matricích, jako je moč, krevní plazma či krevní sérum. Vyvinuté metody a postupy mohou sloužit v klinické praxi k rutinní diagnostice, případně se mohou stát základem pro simulaci metabolomických procesů probíhajících v lidském organismu a ke studiu oxidačního stresu i různých defektů DNA.

The presented thesis is focused on the development and description of analytical methods for the determination of selected biomarkers in clinical diagnostics using electrochemistry as an important element of modern instrumental techniques. Biomarkers are substances whose presence usually indicates various types of serious diseases and conditions, or exposure to harmful chemicals. In this work, attention was paid mainly to the determination of metanephrines, markers of the tumor pheochromocytoma, and indoxyl sulfate, an important marker of several health complications, from renal failure through intestinal dysbiosis to central nervous system disorders. Part of the work is also devoted to the development of an original instrument used for metabolomics research.
Combinations of liquid chromatography with electrochemical and spectrophotometric detector as well as connection of electrochemistry with mass spectrometry were used to analyze selected compounds in complex biological matrices such as urine, blood plasma or blood serum. The developed methods and procedures are higly sensitive, accurate and precise. It is possible to use them in both clinical practice for routine diagnosis, or in clinical research for studying of human metabolomics processes, oxidative stress and DNA defects.
