Bakalářská práce se zaměřuje na virus šarky švestky s důrazem na hostitele mandloň obecnou. Práce je rozdělena do dvou částí. Teoretická část obsahuje literární rešerši shrnující dosavadní poznatky o viru i hostiteli a experimentální část zahrnuje detekci tří významných virů peckovin v celkem 62 stromech, z toho 48 mandloních obecných pocházejících ze 4 oblastí v České republice. Cílem práce je zhodnotit význam mandloně obecné v epidemiologii PPV. Ve vzorcích byly provedeny detekce Plum pox virus (PPV), Prunus necrotic ringspot virus (PNRSV) a Prune dwarf virus (PDV).
Mandloň obecná může být vzácně přirozeně infikována PPV, avšak nevykazuje výrazné symptomy spojené s ekonomickými ztrátami (Festić, 1978), které mohou u jiných náchylnějších hostitelů i úplně znehodnotit úrodu. Symptomy šarky na mandloních nejsou spolehlivým indikátorem přítomnosti PPV. Mohou znamenat infekci jinými patogeny, například PNRSV, PDV či ACLSV.
V současnosti se PPV infikované mandloně nachází ojediněle v Turecku (Akbaş a kol., 2011; Karabacak a Ilba?i, 2011; Ilba?i a Citir, 2014), Maďarsku (Pribék a kol., 2001; Kölber a kol., 2003), Bulharsku (Topchiiska, 1996; Kamenova a Milusheva, 2005) a Rumunsku (Macovei a Diaconu, 2001). Incidence je však nízká a navíc v oblastech, kde převažuje pěstování jiných druhů peckovin. Proto tyto výjimečné případy infekce nejsou závažnou hrozbou tamější ovocné produkce. Mimo Evropu byl PPV na mandloních potvrzen v Čile (Herrera a kol., 1994; 2013).
V souboru testovaných vzorků nebyla zjištěna infekce PPV, přestože některé stromy jevily na listech symptomy možných virových onemocnění. Výsledky potvrdily očekávání, že v současnosti na našem území nejsou mandloně přirozenými hostiteli PPV a nepředstavují hrozbu v jeho epidemiologii. Za vhodných podmínek mohou být hostiteli experimentálními (Dallot a kol., 1997; Damsteegt a kol., 2007a; Petrov a Stoev, 2011). Pouze kmeny PPV-M, -D a -T nyní infikují mandloně (Pribék a kol., 2001; Kölber a kol., 2003; Ilbagi a Citir, 2014). Ve vzorcích však byly detekovány případy jednotlivých i směsných infekcí PDV a PNRSV. PNRSV byl detekován v koinfekci s PDV v pěti ze čtyřiceti mandloní a jednotlivě v pěti višních a jedné švestce. PDV byl detekován ve třiceti ze čtyřiceti osmi mandloní, ve třech švestkách a dvou višních.

The bachelor thesis is focused on on the Plum pox virus with emphasis on host almond. The thesis is divided into two parts. The theoretical part includes literature search which summarizes current knowledge about both the virus and the host. The experimental part of this thesis is focused on detection of 3 different viruses in samples collected from 62 trees, including 48 almonds, originating from 4 different locations in the Czech Republic. The aim of the thesis is to evaluate the importance of almonds in PPV epidemiology. Detections of Plum pox virus (PPV), Prunus necrotic ringspot virus (PNRSV) and Prune dwarf virus (PDV) were carried out in samples.
Almond can rarely be naturally PPV infected, but does not exhibit strong symptoms associated with economic losses (Festić, 1978), which may be present in other susceptible hosts and sometimes completely devaluate the crop. Symptoms of sharka on almonds are not reliable indicator of PPV infection. They may be related to other viral infections, for example by PNRSV, PDV or ACLSV.
Nowadays in Europe, PPV infected almonds are only present in Turkey (Akbaş et al., 2011; Karabacak and Ilba?i, 2011; Ilba?i and Citir, 2014), Hungary (Kölber et al., 2003), Bulgaria (Topchiiska 1996; Milusheva, 2005) and Romania (Macovei a Diaconu, 2001). However, the incidence is low and in areas where other species of stone fruit are predominant. Therefore, these rare cases are not a serious threat to the local fruit production. Out-of-Europe PPV infected almonds has been confirmed in Chile (Herrera et al., 1994; 2013).
In tested samples no PPV infections were detected although some of the trees including almonds had characteristic symptoms of possible viral disease. The results confirmed the expectation that almonds are not a natural PPV host in Czech republic and represent no significant risk in epidemiology of PPV so far. Under suitable conditions almonds may be experimental hosts (Dallot a kol., 1997; Damsteegt a kol., 2007a; Petrov a Stoev, 2011). Only strands PPV-M, -D and -T are now infecting almonds. However, the situation might get worse with changing climatic conditions. In same samples some cases of both individual and mixed infections by PDV and PNRSV were detected. PNRSV was found in coinfection with PDV in five almonds and individually in five tart cherries and one plum. PDV was found in thirty out of fourty-eight almonds, three plums and two tart cherries.
