Course content
|
Preparative chromatographic methods have become indispensable part of experimental practice of organic chemists, biochemists and other chemical experts. The chromatographic methods are more widely used as an effective tools for separation and purifying of organic synthesis products in cases in which classical methods of product separations such as extraction, precipitation, crystallization are not sufficiently effective. Other application of (semi)preparative chromatography could be an isolation of pure substances from complex natural extracts. Today it is becoming commonplace that chemical laboratories are equipped by modern chromatographic instrumentation and it is important advantage for chemist if he is familiar with basic principles and has an experimental preparative chromatography skills. That is why the laboratory practice aims to (1) familiarize students with basic principles of preparative chromatography, (2) teach students to develop an efficient chromatographic method, (3) teach students how to operate chromatographic instrumentation and notify them of the typical troubles associated with instrument use (troubleshooting), (4) new theoretical and practical knowledge will be applied to solve assignments in labs. Students will learn how to use traditional methods such as preparative thin-layer chromatography, column chromatogramy as well as "flash" chromatogram or preparative HPLC system. Attention in the course will be paid to a change from analytical separation conditions to preparative conditions. The practice will take place in the laboratories of the Department of Organic Chemistry UP that are adequately equipped with modern chromatographic instrumentation (analytical chromatography, preparative HPLC, "flash" chromatograph).

The laboratory practice from preparative chromatography methods aims to familiarize students with basic chromatographic techniques that can be applied during purifying of reaction products or separations of other complex mixtures.
