Lectures 1) PCR principle, PCR types, practical application 2) Primers, their design and use, work with sequences and databases 3) Recombinant technologies, restriction, ligation 4) Vector types, cloning 5) Nucleic acid isolation 6) Gene expression quantification I, transcription detection 7) Gene expression quantification II, expression determination 8) Transgenic organisms, transformation 9) Gene editing 10) Application of molecular genetics in breeding The laboratory course will focus on basic molecular biology methods and their applications. 1) Introductory session, preparation of plant material, 4 hours (2/2) 2) Differentiation between two spring barley varieties using microsatellites, PCR, and electrophoresis, 4 hours (2/2) 3) RNA isolation using TRIzol, concentration determination, 4 hours (2/2) 4) RNA quality check, electrophoresis, DNase treatment, 4 hours (2/2) 5) Reprecipitation, reverse transcription of RNA to cDNA, 4 hours (2/2) 6) RT-PCR, electrophoresis, data analysis, 4 hours (2/2) 7) Colloquium, examination, 2 hours

The course introduces students to basic and advanced methods of molecular biology and their practical use. The course focuses on modern laboratory techniques and procedures used to study nucleic acids (DNA, RNA) and gene expression. The course also includes information on methods of plant transformation, including gene editing and their application in genetics and breeding.

Student performance
During the course, students will learn theoretically and, most importantly, practically about molecular biology methods and their use in genetic engineering. They will learn how to design primers and amplify the gene of interest using PCR. |
