Lecture: 1. Lymphatic organs. 1. Cellular mechanisms of natural immunity - (epithelial cells, phagocytosis, granulocytes, monocyte-macrophages, APC, PRR, NK cells). Selected aspects of mucosal immunity 2. Cellular mechanisms of specific immunity - (CD4 - Th1, Th2, Th9, Th17, Tfh, Treg, CD8 T, , gamma-delta T lymphocytes). Humoral mechanisms of specific immunity (antibodies, immunocomplexes). B lymphocytes and their maturation, activation, and population affinity maturation and isotype shift 3. Intercellular communication - cytokines, chemokines, adhesion molecules and other regulatory mechanisms of immunity. Antigen presentation and induction of specific immune responses 4. Major histocompatibility complex, other polymorphic immune genes. HLA in transplantation, autoimmunity, susceptibility to infection. Anti-tumour immunity 5. Mechanisms of immunopathological reactions - hypersensitivity types I-IV. Allergies - nature and manifestations. Autoimmunity and autoimmune diseases - central and peripheral tolerance, mechanisms of its disruption, stages of autoimmune disease. Immunodeficiency primary, secondary Exercise: 1. Laboratory testing of OFI phagocytosis, Burst test 2. Flow cytometry review of principles and applications of classical, spectral and mass cytometry. Phenotypic determination of T and B lymphocyte populations, intracellular cytokine determination, combined examination of surface and intracellular proteins and nucleic acids 3. Investigation of humoral immunity: precipitation methods RID, nephelo, turbidi., solid phase ELISA, ImmunoCap, multiparametric solid phase methods Luminex, ISAC, FEIA, ... Determination of antibodies, complement, acute phase proteins, CIK. Laboratory testing in allergic patient: total and specific IgE, markers of allergic inflammation 4. HLA typing: serological, genotyping PCR SSP, PCR SSO, Sanger, NGS, anti HLA antibodies 5. Laboratory testing of autoantibodies: indirect immunofluorescence, ELISA, Alegria system.

To introduce principles of immunology and involvement of immune system in the etiopathogenesis of infectious diseases, tumors, autoimmune and allergic diseases. To introduce present principles and prospects of immune-based therapy, and finally provide informations about methods of clinical immunology labory investigation and methods experimental immunology.
Students will understand the basics of immunology and the involvement of the immune system in the etiopathogenesis of infectious diseases, tumor growth, autoimmune and allergic diseases. Students will know the basic principles and options for immunologically based therapies and basic techniques and methods of laboratory determination in clinical immunology practice and experimentation. |

Written examination in the form of test questions confirming knowledge of the subject matter is 1 hour long, 25 questions, maximum score 25. A: 25-23 points B: 22-20 points C: 19-17 points D: 16-14 points E: 13-12 points F: less than 12 points |
