Course content
|
The basics of immunochemistry and immunoanalysis. Nomenclature, chemical equilibrium, types of interaction, avidity, immunogen, immune response, immunization, creation of polyclonal antibodies, monoclonal antibodies, recombinant antibodies, isolation and purification of antibodies. Types of imunoanalysis (radioisotopic, enzymatic, fluorescence, chemiluminiscence and others). Heterogenic and homogenic types of analysis, competitive and non-competitive types of analysis, the separation systems, optimization and characterization of immunoanalytical methods. Interpretation of analytical signal, benefits and limitation of immunoanalytical methods. Application of immunanalytical methods in clinical practice and in environmental analysis. Immunoanalysis of low-molecular compounds. Immunoanalytical screening methods. Specifity of low-molecular haptens. The preparation of haptens and preparation of haptens to production of pointing antibodies. The utilization of conjugation techniques. The methods for marks of antigen and antibodies. Conjugation reagent and their preparation, preparation of real samples for immunoanalysis. The matrix effects in immunoanalysis, the precision and validity of real application. Development of enzymatic imunoanalytical techniques for detection and determination of triazine herbicides. Immunochromatography. The connection of immunoanalysis with separation techniques (gel electrophoretic , HPCE, HPLC), imunosemnsors. The progress of immunoanalytical techniques.

Learning outcomes
|
The aim is to learn methods and immunoassay of low-molecular substances
Describe the reaction between antibody and antigen. Define the terms antigenicity, hapten, avidity, immunogen. Describe the composition of immunoglobulins. Apply the theoretical knowledge to immunochemical properties of antibody and antigen to design of immunochemistry based on analytical methods. Apply the knowledge to determination of low molecular compounds (haptens) in many types of samples.

Prerequisites
|
The knowledge of basics of general, organic, anorganic, physical and analytical chemistry is recommended.
