Course content
- Introduction: Chemical compounds as mutagens and carcinogens, mutations, origin and types of mutations - Basic properties of nucleic acids: Chemical composition, structure and properties of nucleic acids. Mechanism of transfer of genetic information and its changes (replication, transcription). Determination of nucleotide sequence of nucleic acids (endonucleases, exonucleases, restriction enzymes, chemical /Maxam-Gilbert/ method, the chain-termination /Sanger/ method) - brief repetition of knowledge from molecular biology and biophysics - Reactions of mutagens, carcinogens and cytostatics: direct reactions, metabolic activation; cytostatics- types of most frequently used cytostatics, determination of targets of their effects. - Repair of damaged DNA in mammalian cells a) total repair b) excision repair : base excision repair, nucleotide excision repair, mismatch repair, c) repair of double strand breaks, d) tolerant repair - Methods used to study reactions of mutagens, carcinogens and cytostatics with DNA: a) DNA binding, detection and characterizations of DNA adducts of carcinogens, sequence preference of DNA binding, b) formation of interstrand cross-links, c) alterations of conformation and stability of DNA d) reactions with cellular proteins and repair of damaged DNA, e) methods and procedures for determination of mutagenicity of chemical compounds.

Prerequisites
Basic knowledge of molecular and general biophysics and molecular biology.
