Course content
It gives an overview on history and basic principles of plant tissue cultures, setups for aseptic work, totipotency of plant cell, regeneration potential of plants, somatic variability and its utilization, morphogenesis (organogenesis, embryogenesis). The basic methods of plant tissue culture techniques (meristem culture, callus culture, anther culture, embryo rescue, protoplast isolation and fusion). Utilization methods in practise (plant breeding, plant propagation, transgenic plants, secondary metabolites, in vitro gene bank, in vitro phytopathology, in vitro selection to abiotic stresses, in vitro toxicology). Practical exercise: aseptic work, obtaining of practical experiences on techniques of plant tissue cultures -micropropagation, virus-free plants, embryo rescue, anther culture, protoplast isolation and somatic hybridization and transfer of plants to the substrate (non-sterile conditions). For the time required of the methods - practises are blocked.

Learning outcomes
This course concurs and axpands to lectures of general botany and plant physiology. Practical exercises are focused on work in aseptic environment and manipulation of plant materiál.
Student should be able to (after attending the course): - manage work in aseptic conditions - explain regeneration abilities of plants - apply basic methods of in vitro cultures used in Plant breeding and biotechnologie; gene banks, in vitro phytopathology - predict potential and perspectives of Explantate Culture Techniques - set up and evaluace the experiments - dokument the experiments - to work in a team (group)
Prerequisites
After graduating student is able to understand the problems of plant tissue cultures, methods and of their use in practice.

Assessment methods and criteria
Dialog

Participation in practices (70%). Verbal colloquium in work groups in extent of the lectures and results of the practices (ca. 1h).
